Resumo O estudo da diversidade genética de Theobroma speciosum é importante, pois pode ser utilizado em programas de melhoramento, uma vez que a espécie está intimamente relacionada a espécies de grande valor econômico, como Theobroma cacao (cacau) e Theobroma grandiflorum (cupuaçu). Assim, o objetivo deste trabalho é caracterizar a diversidade genética intra e interpopulacional de Theobroma speciosum em populações naturais na Amazônia brasileira. Foram selecionados e genotipados 90 indivíduos de T. speciosum provenientes de quatro populações situadas em diferentes estados da Amazônia Legal. Os dados foram obtidos por meio da análise de microssatélite e a diversidade genética foi caracterizada através do número de alelos, número de alelos privados, índice de fixação e heterozigosidade observada e esperada. A análise bayesiana, a AMOVA e PCOa foram utilizadas para revelar a estrutura genética molecular das populações, através dos programas Structure e GenAIEx 6.5, respectivamente. Todas as populações estudadas apresentaram níveis de diversidade gênica, contudo, as populações AUR, API e MAC apresentaram grande similaridade, enquanto a população de Rio Branco apresentou maior heterozigosidade e menor endogamia do que as outras, se tornando um ponto de refúgio na Amazônia, e a mais importante população para a conservação de T. speciosum.

Abstract The genetic diversity of Theobroma speciosum is important because its use in breeding programs, once the species is closely related to species of great economic value such as Theobroma cacao (cocoa) and Theobroma grandiflorum (cupuaçu). Thus, the objective of this work is to characterize the intra and interpopulational genetic diversity of Theobroma speciosum in natural populations in the Brazilian Amazon. Ninety individuals of T. speciosum from four populations localized in different states of legal Amazon were selected and genotyped. The data were obtained by fluorescence microsatellite analysis and the number of alleles, number of private alleles, fixation index, observed and expected heterozygosity were analyzed. Bayesian analysis, AMOVA and PCOa were used to reveal the molecular genetic structure of the populations, using the programs Structure and GenAIEx 6.5, respectively. All populations studied present great levels of gene diversity, although, there was a greater similarity among the AUR, API and MAC populations, while RBC population presented higher heterozygosity and less inbreeding than the others, becoming a possible refuge area in the Amazon, and the most important population for T. speciosum conservation.

Abstract In view of the economic and environmental importance of Bertholletia excelsa Bonpl. for the Amazon and the reducing natural habitat area of this species, the present study proposes to examine genetic variability among accessions of Brazil nut from the Amazon rainforest. Seventeen native Brazil nut genotypes were sampled from Sinop-MT and the genomic DNA were amplified using 15 inter-simple sequence repeat (ISSR) markers. The percent polymorphism and the polymorphism information content (PIC) of each primer were determined. The primers amplified 84.62% of the polymorphic bands and 15.38% of the monomorphic bands. The PIC for each primer ranged from 0 to 0.68. The Unweighted Pair Group Method with Arithmetic Mean (UPGMA) was the most efficient in group representation, as it showed the highest cophenetic correlation coefficient (CCC = 0.92), the lowest stress (12.31%) and the lowest distortion (1.51%). The use of ISSR markers was an efficient tool in the study of genetic diversity among Brazil nut genotypes, and the genetic diversity found can be used for conservation and pre-breeding programs for this crop.

RESUMO A ocorrência da correlação negativa entre produtividade de grãos (PG) e capacidade de expansão (CE) no milho-pipoca contribui para dificultar o processo seletivo. No intento de contornar esse inconveniente, foi proposto, o caráter de importância, volume por pipoca expandida por hectare (VP). O objetivo deste estudo foi desenvolver o nono ciclo da população UENF-14 de milho-pipoca, estimar o efeito direto e indireto de VP e averiguar o progresso genéticos entre todos os ciclos seletivos. Duzentas progênies endogâmicas foram obtidas e avaliadas em blocos casualizados, com arranjos em repetições dentro de sets, sendo nove sets, oito constituídos por 25 progênies e um set com todos ciclos anteriores (oito) com três repetições. Avaliou-se a altura média da planta, prolificidade, peso de cem grãos, produtividade de grãos, capacidade de expansão e volume por pipoca expandida por hectare. Realizou-se a análise de trilha para desdobrar os efeitos diretos e indiretos e o índice de seleção Mulamba e Mock para seleção de quarenta progênies superiores. Houve variabilidade genética entre as progênies para todas características. Existe relação de causa e efeito entre o VP com a PG e CE, permitindo ganhos simultâneos com a seleção indireta. A seleção das progênies superiores com a utilização do peso atribuído por alternativas proporcionou maiores ganhos preditos de VP (20,73%). As médias obtidas em todos os ciclos indicaram aumentos, demonstrando a eficiência do uso da seleção recorrente em programas de melhoramento de milho-pipoca.

ABSTRACT The occurrence of negative correlations between grain yield (GY) and popping expansion (PE) in popcorn complicates the selection process. In an attempt to overcome this inconvenience, we proposed the use of the importance characteristic volume per popcorn per hectare (PV). The objectives of this study were to develop the ninth cycle of the UENF-14 popcorn population, to estimate the direct and indirect effects of PV and to verify the genetic progress among all selective cycles. We obtained and evaluated 200 inbred progenies in randomised blocks arranged in replicates within sets, with nine sets consisting of 25 progenies and one set with all previous eight cycles with three replicates. The average height of the plants, prolificacy, 100-grain weight, grain yield, grain popping expansion and volume per popcorn per hectare were evaluated. Track analysis was performed to determine the direct and indirect effects, and the Mulamba and Mock selection index was calculated for the selection of the 40 superior progenies. There was genetic variability among progenies in all evaluated traits, with a cause-and-effect relationship between PV with GY and PE, allowing simultaneous gains with indirect selection. The selection of the best progenies was more effective when using arbitrarily assigned weights, which provided higher predicted gains for PV (20.73%). The means obtained in all cycles indicated increases, demonstrating the efficiency of using continuous recurrent selection in popcorn breeding programs.

ABSTRACT The present study resulted from the lack of information on the genotypic expression ruling the resistance to rust and on the main traits of economic importance to the herein addressed crop. The combining ability of 16 popcorn lines and hybrids was assessed in partial diallel cross in two representative locations in Rio de Janeiro State, Brazil, where forty simple hybrids were obtained. The experiments followed a completely randomized block design with four repetitions and were conducted under natural infestation conditions in order to test the southern rust severity in the entire plant (SSW) and in the leaf of the first ear (SFPE), as well as the morpho-agronomic traits of greater interest: grain yield (GY) and popping expansion (PE). Additive gene effects prevailed in PE and the non-additive ones stood out in GY; however, both gene effects were important to the southern rust resistance characteristics (SSW and SFPE). Hybrids L65 × P10 and P7 × L70 presented appropriate specific combining ability (SCA) values in all traits; therefore, they were promising for the selection of superior genotypes. Lines L80, L76 and L77 expressed negative general combining ability (GCA) values in SSW and SFPE; line P10 recorded high and positiveGCA values in GY and PE.

ABSTRACT Compared with the common corn, popcorn shows greater susceptibility to pests and diseases occurrence, being the use of resistant cultivars the most efficient strategy. In this regard, Germplasm Banks deserve special attention because they contain accessions that can be used as sources of resistance in breeding programs. The State University of Northern Rio de Janeiro (UENF) maintains a popcorn Germplasm Bank with accessions from tropical and temperate countries, including Diversity Centers for the species. In this study, we investigated the performance of 37 popcorn accessions landraces, hybrids, and inbreed lines ― from the Germplasm Collection of UENF’s Popcorn Breeding Program with respect to the fungal leaf diseases northern leaf blight (NLB) and southern leaf blight (SLB), caused by Exserohilum turcicum and Bipolaris maydis, aiming to select promising accessions for genetic resistance to be used in future crosses to generate new resistant cultivars for farmers. The dendrogram scattering by the UPGMA multivariate technique was efficient in discriminating resistant accessions. Satisfactory results were obtained for resistance to NLB in accessions L71, L75, L76, P7, and PARA-172, which can thus be considered remarkable sources of resistance. For resistance to SLB, the superior accessions that can be indicated as sources of resistance are ARZM-05083, ARZM-07049, and PARA-172.

RESUMO. O êxito de qualquer programa de seleção recorrente depende da variabilidade genética da população avaliada, sendo utilizada para se referir à diversidade de alelos existentes nos vários locos genéticos. Assim, o objetivo do presente estudo foi investigar o impacto da seleção recorrente em nove ciclos da população UENF-14 de milho-pipoca através da análise da diversidade genética e da estrutura utilizando marcadores microssatélites (SSR-ESTs). O DNA genômico foi extraído de folhas jovens de 25 indivíduos de cada ciclo (C0, C1C2, C3, C4, C5, C6, C7 e C8) totalizando 225 amostras da população UENF-14. Para a análise da diversidade genética entre os ciclos de seleção recorrentes foram utilizados cinquenta marcadores SSR-ESTs, dos quais 16 revelaram ser polimórficos. Trinta e quatro alelos foram detectados, com uma média de 2,13 alelos por locos. Ao longo de todos os ciclos de seleção recorrentes houve uma redução na heterozigosidade e aumento na endogamia. A estrutura da população mostrou um compartilhamento de alelos inferindo que alguns podem ser direcionados para as principais características de seleção.

ABSTRACT. The success of any recurrent selection program depends on the genetic variability of the evaluated population, which is used to refer to the diversity of existing alleles at many genetic loci. Thus, the goal of the present study was to investigate the impact of recurrent selection across nine cycles of a UENF-14 popcorn population through the analysis of genetic diversity and structure using microsatellite markers (EST-SSRs). Genomic DNA was extracted from young leaves of 25 individuals from each cycle (C0, C1 C2, C3, C4, C5, C6, C7, and C8), totaling 225 samples from the UENF-14 population. Fifty EST-SSR markers were used for the analysis of genetic diversity across the recurrent selection cycles, 16 of which were polymorphic. Thirty-four alleles were detected, with an average of 2.13 alleles per locus. Throughout all the recurrent selection cycles, there was a reduction in heterozygosity and an increase in inbreeding. The population structure showed a sharing of alleles, inferring that some may be directly related to the main selection characteristics.

RESUMO. Com o objetivo de investigar a resistência a Exserohilum turcicum na população de milho-pipoca UENF-14, bem como inclusão desta característica na análise, duzentas e dez famílias de meios-irmãos foram avaliados usando delineamento em blocos incompletos, com tratamentos dispostos em repetições dentro "Sets". Em relação à fonte de variação Famílias dentro "Set" (F / S), foi encontrada variabilidade genética suficiente para ser explorada nos próximos ciclos do programa de melhoramento de milho-pipoca da UENF em relação à doença helmintosporiose. A variedade de polinização aberta UENF-14 tem na sua base genética fonte de resistência à doença, devido ao fato que a mesma se originou a partir de cruzamento com a cultivar de grãos amarelos Americana, que é resistente à doença. Tais resultados vêm reforçar a possibilidade de seleção de famílias resistentes através dos ciclos. Portanto, concluiu-se que a seleção foi eficaz em manter o equilíbrio da doença na população em estudo e que a fonte de resistência não é perdida com o avanço dos ciclos de seleção. A adição da variável doença foliar na análise é de extrema importância para o melhoramento do milho-pipoca, pois torna possível agregar os genes para resistência a esta doença juntamente com as características agronômicas de interesse.

ABSTRACT . To investigate the genetic resistance to northern corn leaf blight (Exserohilum turcicum) disease in the UENF-14 popcorn population and the inclusion of this trait in the analysis, two hundred ten popcorn half-sib families were evaluated using an incomplete block design with treatments arranged in replicates "Sets". Regarding the Families within "Set" (F/S) source of variation, enough genetic variability was found for northern corn leaf blight disease resistance to be explored in the next cycles of the UENF popcorn breeding program. The open-pollinated UENF-14 variety has a source of resistance to this disease in its genetic basis, given that it originated from crossing with an American cultivar with yellow grains that is resistant to disease. These results strengthen the possibility of selecting resistant families across the cycles, so it is concluded that the selection was effective at maintaining the balance in the occurrence of the disease in the study population and that the source of resistance is not lost with advancing cycles. The addition of the foliar disease variable in the analysis is of the utmost importance for the improvement of popcorn as it makes it possible to aggregate genes for resistance to this disease along with agronomic traits of interest.

Abstract This study aimed to evaluate the effect of recurrent selection on the genetic variability of UENF-14 population after six selections. Two hundred and ten half-sib families were evaluated in two environments in the state of Rio de Janeiro, using incomplete randomized blocks design with treatments arranged in replication within "Sets". There was significant effect for Families within the "Set" (F)/S, proving that there is enough genetic variability to be exploited in the popcorn breeding program of UENF. The significance for the source of variation Environment (E) shows that the environments were distinct enough to promote differences between the evaluated characteristics. It was found that for both characteristics of greatest interest, GY and PE, the magnitude of the additive variance remains with close values in advanced cycles of UENF-14 population, indicating that variability remains, with no evidence of decreases in advanced cycles. This is concluded by the longevity of UENF breeding program.

RESUMO O uso de topcross vem se mostrando uma opção interessante para a cultura do milho; entretanto, para o milho-pipoca, há pouca informação sobre a escolha do testador adequado. Nesse contexto, este estudo teve como objetivo analisar quatro testadores, sendo duas variedades de polinização aberta (BRS Angela e UENF-14), um híbrido topcross (IAC 125) e uma linhagem (P2), visando avaliar a capacidade combinatória de 50 famílias S3 de milho-pipoca, obtidas a partir da variedade UENF-14. Avaliou-se o rendimento de grãos (RG) e capacidade de expansão (CE). Obtiveram-se as estimativas das capacidades geral e específica de combinação, bem como efetuou-se a discriminação de cada testador por meio de índice de diferenciação. Os testadores BRS Angela (para RG) e IAC 125 (para CE) foram os que se destacaram, quando combinados com as melhores famílias S3derivadas da variedade UENF-14, para produção de híbridos de milho-pipoca para as Regiões Norte e Noroeste Fluminense.

ABSTRACT The use of topcross has proven to be an interesting option for the maize crop; however, for the popcorn, there is little information about the choice of the appropriate tester. In this context, this study aimed to analyze four testers including two open pollinated varieties(BRS Angela and UENF-14), one topcross hybrid (IAC125) and a line (P2), to evaluate the combining ability of 50 S3 families of popcorn, obtained from the UENF-14 variety. Popcorn families were evaluated for grain yield (GY) and popping expansion (PE). The estimates of general and specific combining abilities were obtained and the discrimination of each tester through differentiation index was carried out. The testers BRS Angela (for GY) and IAC 125 (for PE) were the most adequate, when combined with the best S3 families derived from UENF-14, for the production of popcorn hybrids for the Northern and Northwestern Fluminense Regions.

Este trabalho objetivou caracterizar acessos de C. canephora oriundos de cultivos comerciais e bancos de germoplasma brasileiros. Um total de 130 acessos das coleções do IAC (São Paulo), UFV (Minas Gerais) e coletados em plantios comerciais no Espírito Santo e Rondônia foram genotipados com 20 novos microssatélites. Métodos multivariados foram utilizados para estimar a relação entre os acessos. Foi observado alto nível de polimorfismo e dois grupos foram identificados: o primeiro foi constituído pelos genótipos conservados nas coleções de germoplasma do IAC e UFV e o segundo foi composto pelos acessos coletados em plantios comerciais. Os acessos do Espírito Santo e de Rondônia formaram dois subgrupos distintos. Apesar do grande polimorfismo encontrado nas lavouras brasileiras de café canéfora, incrementar essa diversidade é necessário, pois um novo limiar de ganhos genéticos é esperado nos programas de melhoramento com a intensificação do uso do germoplasma conservado.

This work aimed to characterize accessions that represent the C. canephora germplasm conserved and cultivated in Brazil. A total of 130 accessions from germplasm banks of IAC (São Paulo), UFV (Minas Gerais) and also collected in plantations of the State of Espírito Santo and Rondônia were evaluated with a set of 20 new microsatellite primers. Multivariate methods were used to estimate the relationship among the accessions. High level of polymorphism and two major diversity clusters were identified. First cluster was composed by the accessions conserved in the IAC and UFV collections and the second was formed by accessions collected in areas under cultivation. Accessions from Espírito Santo and Rondônia were clear separated, composing two subclusters. Despite the great polymorphism found in Brazilian plantations, the diversity may be increased, because a new threshold in the genetic gains is expected on breeding programs with the intensification of the use of conserved germplasm.

O objetivo deste trabalho foi comparar quatro índices de seleção e o método REML/Blup na avaliação de ganhos genéticos preditos das características de interesse ao programa de melhoramento do milho-pipoca UENF 14. Foram avaliadas 200 progênies de irmãos-completos, em uma safra, em dois ambientes (regiões Norte e Noroeste do Estado do Rio de Janeiro), com duas repetições, o que totalizou 800 observações. Avaliaram-se as características altura média de plantas, altura média de inserção da primeira espiga, estande final, tombamento, diâmetro de colmo, prolificidade, rendimento e capacidade de expansão dos grãos. Entre os índices de seleção testados, o de Mulamba & Mock proporcionou os melhores resultados para a seleção das progênies de irmãos-completos. O método REML/Blup mostrou-se muito eficiente, tendo selecionado progênies com desempenhos relativos elevados e com ganhos genéticos preditos melhores que os dos índices de seleção testados. As progênies selecionadas com uso do método REML/Blup, para maior rendimento de grãos, não são as mesmas selecionadas para maior capacidade de expansão. O índice de Mulamba & Mock e o método REML/Blup proporcionam os melhores ganhos preditos para a população UENF 14.

The objective of this work was to compare four selection indices and the REML/Blup method in the evaluation of the predicted genetic gains for the characteristics of interest to the breeding program of the UENF 14 popcorn. Two hundred full-sib progenies were evaluated in one season, in two locations (North and Northwest Regions of the state of Rio de Janeiro, Brazil), with two replicates, totaling 800 observations. The characteristics evaluated were average plant height, average height of first ear insertion, final stand, tipping, stem diameter, prolificacy, grain yield and popping expansion. Among the selection indices tested, Mulamba & Mock's provided the best results for selection of full-sib progenies. The REML/Blup method proved to be very efficient, selecting progenies with high relative performance and better predicted genetic gains than those of the selection indices tested. The progenies selected using the REML/Blup method, for highest grain yield, are not the same as those selected for higher popping expansion. The Mulamba & Mock index and the REML/Blup method provide the best predicted gains for the UENF 14 population.

O presente trabalho tem como objetivo informar a comunidade científica os aspectos relacionados à cultivar de milho-pipoca UENF 14, desenvolvida a partir de cinco ciclos de seleção recorrente da população UNB-2U. A cultivar obteve produtividade de 3047.58 kg ha-1 e capacidade de expansão de 35.69 mL g-1.

The aim of this work is to present the features of the popcorn cultivar UENF 14, developed from five cycles of recurrent selection of the population UNB-2U, to the scientific community. The cultivar produces yields of 3047.58 kg ha-1 and has popping expansion of 35.69 mL g-1.

Marcadores moleculares AFLP, RAPD e SSR foram usados para estudo da diversidade e estrutura genética do germoplasma de Híbrido de Timor. As análises de coordenadas principais, de agrupamento UPGMA baseado na dissimilaridade genética de Jaccard, de agrupamento baseado no modelo Bayesiano, a percentagem de locos polimórficos, o índice de Shannon e a diversidade genética de Nei foram utilizadas para acessar a diversidade genética. As análises demonstraram alta diversidade genética entre os acessos de Híbrido de Timor. A análise de agrupamento UPGMA e o modelo Bayesiano permitiram a alocação dos acessos em três grupos. A estrutura genética dos acessos de Híbrido de Timor foi descrita. A manutenção e manuseio da variabilidade do germoplasma de Híbrido de Timor e seu potencial uso nos programas de melhoramento de cafeeiro foram discutidos.

AFLP, RAPD and SSR molecular markers were used to study the genetic diversity and genetic structure of the Híbrido de Timor germplasm. The principal coordinate analysis, UPGMA cluster analysis based on genetic dissimilarity of Jaccard, Bayesian model-based cluster analysis, percentage of polymorphic loci, Shannon's information index and Nei gene diversity were employed to assess the genetic diversity. The analyses demonstrated a high genetic diversity among Híbrido de Timor accessions. UPGMA and Bayesian cluster analyses grouped the accessions into three clusters. The genetic structure of Híbrido de Timor is reported. The management of Híbrido de Timor germplasm variability and its potential use in breeding programs are discussed.

No estudo da diversidade genética entre e dentro de populações de café, foram usados marcadores EST-SSR, visando avaliar seu potencial para identificar cultivares comerciais e assistir programas de melhoramento. Os 17 marcadores utilizados foram desenvolvidos a partir das seqüências ESTs do Projeto Brasileiro do Genoma Café. Em todos os marcadores observou-se polimorfismo entre os genótipos avaliados, com um número médio de 5,1 alelos por primer. Os maiores polimorfismos foram constatados dentro de C. canephora (88,2%) e em variedades resistentes à ferrugem (35,3%). Dos marcadores analisados, 29,4% distinguiram C. arabica dos Híbridos de Timor (HDT), sendo possível identificar os mais próximos e os mais distantes de C. arabica . A análise dos genótipos agrupados por população revelou diversidade genética de 64% entre populações e 36% dentro delas, com um índice de diferenciação de 0,637. Em seis marcadores distinguiram-se quatro variedades resistentes à ferrugem, constando seu potencial para fingerprint. Esses resultados demonstram a utilidade dos EST-SSR para orientações dos cruzamentos e estudos de diversidade, introgressão e mapeamento genético.

EST-SSR markers were used to investigate the genetic diversity among and within coffee populations, to explore the possibility of their use for fingerprinting of cultivars and to assist breeding programs. Seventeen markers, developed from ESTs (Expressed Sequence Tags) from the Brazilian Coffee Genome Project, were used. All markers showed polymorphism among the genotypes assessed. The average number of allele per primer was 5.1. The highest polymorphisms were found within C. canephora (88.2%) and rust-resistant varieties (35.3%). About 29.4% of the markers differentiated C. arabica from Híbrido de Timor; it was also possible to identify those closest and farthest from C. arabica . The analysis of population-grouped genotypes revealed a 64.0% genetic diversity among and a 36.0% genetic diversity within populations. The differentiation index was 0.637. Six markers distinguished four rust-resistance varieties, showing their fingerprinting potential. These results demonstrate the usefulness of EST-SSR markers for cross orientation, in diversity and introgression studies, and in genetic mapping.
